Detection of scientifically relevant RNAs by Next-Generation RNA Sequencing (RNA-Seq) is greatly limited by highly abundant ribosomal RNAs (rRNAs) which occupy > 90% RNA-Seq reads. Current rRNA depletion solutions however are often costly and limited to well-studied species. 

Ribosomal RNA depletion pools (riboPOOLs) by siTOOLs Biotech is the market's most flexible solution in rRNA depletion. Composed of high complexity pools of optimally-designed biotinylated DNA probes, riboPOOLs are highly efficient, even outperforming Ribo-Zero (Illumina), depleting ~10% more rRNA while leaving other RNA intact.

rRNA Depletion workflow


Composed of a highly complex mixture of optimally-designed 3'-biotinylated DNA probes, riboPOOLs hybridize efficiently and specifically to ribosomal RNAs (rRNAs) present in total RNA. 

Streptavidin-coated magnetic beads are added that allow efficient separation of riboPOOL-bound rRNAs through the high affinity biotin-streptavidin interaction.

Remaining relevant RNAs are purified and concentrated by either ethanol precipitation, spin-column purification or SPRI bead-based clean-up, before analysis by Next-Generation Sequencing, microarray or PCR.
