riboPOOLs are high complexity pools of 3'-biotinylated single-stranded DNA.
riboPOOLs hybridize selectively to rRNA, enabling their removal using streptavidin-coated magnetic beads.
Composed of a highly complex mixture of optimally-designed 3'-biotinylated DNA probes, riboPOOLs hybridize efficiently and specifically to ribosomal RNAs (rRNAs) present in total RNA.
Streptavidin-coated magnetic beads are added that allow efficient separation of riboPOOL-bound rRNAs through the high affinity biotin-streptavidin interaction.
Remaining relevant RNAs are purified and concentrated by either ethanol precipitation, spin-column purification or SPRI bead-based clean-up, before analysis by Next-Generation Sequencing, microarray or PCR.
